چکیده

3,4-Diaminopyridine (3,4-DAP) and its phosphate form, 3,4-DAPP have been used efficiently in the past years to treat muscular weakness myasthenic syndromes with neuromuscular junctions (NMJs) impairment. Pompe disease (PD), an autosomal recessive metabolic disorder due a defect of lysosomal enzyme α-glucosidase (GAA), presents some secondary symptoms that are related transmission dysfunction, resulting endurance strength failure. In order evaluate whether could beneficial effect on this pathology, we took advantage transient zebrafish PD model previously generated characterized. We investigated presynaptic postsynaptic structures, NMJs at electron microscopy level, behavior, before after treatment 3,4-DAPP. After drug administration, observed increase number acetylcholine receptors increment percentage normal structure amelioration embryo recovery typical movements were lost model. Our results revealed early NMJ impairment improvement administration 3,4-DAPP, suggesting potential use as symptomatic patients disease.

Peripheral nerve and neuromuscular junction pathology in Pompe disease.

Pompe disease is a systemic metabolic disorder characterized by lack of acid-alpha glucosidase (GAA) resulting in ubiquitous lysosomal glycogen accumulation. Respiratory and ambulatory dysfunction are prominent features in patients with Pompe yet the mechanism defining the development of muscle weakness is currently unclear. Transgenic animal models of Pompe disease mirroring the patient phenot...

Neuromuscular junction in health and disease.

A number of illnesses and other factors can affect the function of the neuromuscular junction (NMJ). These may have an affect at pre- or post-junctional sites. This review outlines the anatomy and the physiology of the NMJ. It also describes the mechanisms and physiological basis of many of the disorders of the NMJ. Finally, the importance of these disorders in anaesthetic practice is discussed.

3,4-Diaminopyridine may improve neuromuscular block during botulism

In September 2011, two outbreaks of botulism toxemia (toxin A) incriminating the same artisanal product were responsible for nine cases of botulism in France. Among these cases, three women (Cases 1, 2, and 3) aged 27, 29, and 23 years, respectively, were hospitalized in our ICU for mechanical ventilation support. They rapidly received a trivalent botulism antitoxin serum. Two weeks later, Case...

Neuromuscular Junction Effects Does Pb Produce Neuromuscular Junction Changes?

Acetylcholinesterase (AChE) inhibitors, including PB, produce acute destructive changes at the neuromuscular junction, the site of connection between nerve cells and muscle fibers, at which nerve cells signal muscles to contract. Because studies of the neuromuscular junction require microscopic inspection of muscle tissue and tests of electrical and chemical properties of the neuromuscular junc...

Feasibility of DBS screening to identify adult patients with Pompe disease in a neuromuscular clinic population

Introduction In January 2012, a program was set up to rapidly screen patients with undiagnosed muscle disorders who were referred to a neuromuscular specialist in an adult hospital. A nurse was trained to do the DBS procedure as part of a series of required laboratory tests. All individuals referred to a neuromuscular specialist, either within the context of a neuromuscular clinic or an EMG lab...
